I'm upgrading so I can start using Adobe Encore with my edited movies.
Does anyone know if Premiere 6.5 and Premiere Pro co-exist on one PC?
Or must I upgrade one to the other and then start using Pro from then
    on?

They work fine on same machine.
They install in different directories and can even both be running at same
    time.
However don't try to capture or export to tape from both at the same time!!

  3. What if you have plug-ins, such as Inscriber or Graffiti, or capture
    cards such as the Matrox?

I don't know Inscriber or Graffiti but unless they are both hitting a piece
of hardware at the same time and both versions of Premiere are running and
using Graffiti or Inscriber they should work.

Some capture cards may come alive when Premiere runs even if you are not
capturing in which case running both versions AT THE SAME TIME might not
    work.
