    Hi,
I have an image file with two color. eg, red and black, how do I can
separate these two colors in two layers, I mean that one layer is all black
    and other is all red.

    Thanks,

Have you tried Select/ Color Range?

With colors so distinct, the magic wand may work well. Cut and paste one
into a new layer and the other will be all alone in the old one.
I'm not sure how the colors are laid out, so you might want to uncheck
    "contiguous."
